et al., 2010) and bind to CArG-boxes, which are small 8–10 bp DNA sequences composed of 6–8 A or Ts flanked by C and G bases (de Folter and Angenent, 2006). The Arabidopsis genome contains an estimated 340,000 potential MADS binding sites (de Folter and Angenent, 2006), leading to the intriguing question: What is the biological significance of the large number of potential MADS binding sites, and how many target genes do MADS proteins really regulate? Recently, several studies reported on the genome-wide identification of target genes of MADS proteins using chromatin immunoprecipitation (ChIP) followed by sequencing (ChIP-seq) or microarray analysis (ChIP–chip; Kaufmann et al., 2009, 2010; Zheng et al., 2009; Deng et al., 2011). Based on these studies, it appears that MADS proteins may directly regulate thousands of target genes (Figure 1A), and may serve to integrate different biological processes. Molecular modeling and expression analysis of a MADS-box cDNA from mango (Mangifera indica L.)
MADS-box genes are a large family of transcription factors initially discovered for their role during development of flowers and fruits. The MADS-box transcription factors from animals have been studied by X-ray protein crystallography but those from plants remain to be studied. In this work, a MADS-box cDNA from mango encoding a protein of 254 residues was obtained and compared. Based on phylo...
متن کاملRegulation of miR159 and miR396 mediated by Piriformospora indica confer drought tolerance in rice
Drought stress is one of the most determinative factors of agriculture and plays a major role in limiting crop productivity. This limitation is going to rising through climate changes. However, plants have their own defense systems to moderate the adverse effects of climatic conditions. MicroRNA-mediated post-transcriptional gene regulation is one of these defense mechanisms. The root endophyti...
متن کاملA MADS box protein interacts with a mating-type protein and is required for fruiting body development in the homothallic ascomycete Sordaria macrospora.
MADS box transcription factors control diverse developmental processes in plants, metazoans, and fungi. To analyze the involvement of MADS box proteins in fruiting body development of filamentous ascomycetes, we isolated the mcm1 gene from the homothallic ascomycete Sordaria macrospora, which encodes a putative homologue of the Saccharomyces cerevisiae MADS box protein Mcm1p. Deletion of the S....
متن کاملDNA-binding specificity of Mcm1: operator mutations that alter DNA-bending and transcriptional activities by a MADS box protein.
The yeast Mcm1 protein is a member of the MADS box family of transcriptional regulatory factors, a class of DNA-binding proteins found in such diverse organisms as yeast, plants, flies, and humans. To explore the protein-DNA interactions of Mcm1 in vivo and in vitro, we have introduced an extensive series of base pair substitutions into an Mcm1 operator site and examined their effects on Mcm1-m...
متن کاملFunctional interactions between the Forkhead transcription factor FOXK1 and the MADS-box protein SRF
The combinatorial control of gene expression by the association of members of different families of transcription factors is a common theme in eukaryotic transcriptional control. The MADS-box transcription factors SRF and Mcm1 represent paradigms for such regulation through their interaction with numerous partner proteins. For example, in Saccharomyces cerevisiae, Mcm1 interacts with the forkhe...
